Boston-based Samuels & Assoc. has begun demolition work to prepare for construction of 201 Brookline Ave., a 500,000-square-foot office-lab tower. 

Venture capital firm Third Rock Ventures will move its East Coast headquarters from Newbury Street into 24,000 square feet. One of its portfolio companies, Cambridge-based Tango Therapeutics, will more than double its footprint after leasing 65,000 square feet at 201 Brookline Ave. 

Designed by Elkus Manfredi Architects, the 14-story tower will include 40,000-square-foot floor plates, lab-ready infrastructure and over 14-foot floor-to-floor heights. 

Since 2007, Third Rock Ventures has raised $2.7 billion for its investments in more than 50 life science companies, including a $55 million series A round for Tango Therapeutics in 2017.
